First, construct a standard curve of migration vs. fragment size for the standards. The size of each unknown band can then be determined from the standard curve. Note that the undigested control pUC19 will not consist of linear DNA fragments. DNA from Blue colony #1 was not digested in the experiment, and looks just like undigested pUC19. DNA from blue colony #2 was successfully cleaved, and looks like pUC19. DNA from white colony #1 was successfully digested and seems to contain the vector and desired target fragment. DNA from white colony #2 looks just like pUC19, even though the colony was white. There are several possible explanations for this observation; for example, we may have identified a pUC plasmid with a mutation in the laca gene.
