Question 7-1.

      The acidic side chains of the protein must be ionized in order for the protein to be attracted to the resin.

Question 7-2.

    The sample must be applied to the ion-exchange column in low salt, since the salt will bind to the available sites on the column and prevent the protein from binding. See figure 4-11 on page 111. Indeed, we will use salt to elute the protein from the column.
