I am investigating the autonomic, visual and olfactory disorders found in early Parkinson’s disease utilizing clinical evaluations coupled with positron emission tomography (PET) studies of nigrostriatal presynaptic monoaminergic innervation utilizing [11C]DTBZ and PET studies of postganglionic sympathetic cardiac innervation utilizing [11C]PMP. Additional investigations are focused on multiple system atrophy, including the natural history of progression, the efficacy of treatment in a pilot study, and the post-ganglionic sympathetic innervation of the heart utilizing PET with [11C]HED. Another set of investigations concerns Alzheimer's disease, testing the hypothesis that in the early stages of the disease, the larger the area of cortical hypometabolism the faster the rate of progression of the disease. This study utilizes PET with [18F]FDG, [11C]DTBZ, and [11C]PIB. In studies with Elan and Wyeth pharmaceutical companies, I am serving as Chair of the Safety Monitoring Committee for passive immunotherapy for Alzheimer's disease. I am currently analyzing the results of a Phase II investigation of Bapineuzumab, a monoclonal antibody directed against deposits of beta-amyloid in the brain of patients with mild to moderate Alzheimer’s disease. |
